A goal from Chris McCann in added on time gave Burnley the three points at Turf Moor to end Blues' three game unbeaten run.
The defender rose unmarked four yards out to head in from an Alan Mahon corner as Town paid for one moment of slackness.
Alan Lee and Simon Walton both went close for Blues and Jim Magilton's side did enough to take something from the game against a side sitting in the top six in the Championship.
The Town boss made three changes to the team that drew at Plymouth, Gary Roberts coming in for his first start, Richard Naylor lining up at the back and Mark Noble returning to the side. The suspended Fabian Wilnis, the injured Billy Clarke and Jaime Peters making way.
It was Blues who dominated the first 20 minutes, Roberts shooting wide with the first effort of the game then Noble's overhead kick cleared by Michael Duff.
Micah Hyde was yellow carded for a foul on Walton as Burnley struggled to get a foothold in the game, with Lee having the clearest chance of the first-half for Blues, going through in a one on one with Brian Jensen but the angle didn't help the striker and his shot was blocked by the Clarets' 'keeper.
Shane Supple was virtually redundant until called upon to deal with a Wade Elliott free-kick but Burnley had a great chance to open the scoring on 25 minutes. The ball fell to McCann eight yards out but Walton did brilliantly to make a brave block.
Steve Jones tested Supple from an acute angle but the Town 'keeper coped easily and a minute later was quick off his line to head clear, Andy Gray penalised for catching the teenager as he challenged for the ball.
James O'Connor was booked for a foul on Roberts on 36 minutes, Lee then spinning brilliantly and firing just wide.
Alex Bruce picked up his ninth yellow card of the season, harshly perhaps, after a foul on Jones as an evenly contested first-half came to an end.
Neither side made any changes at the break with Town maintaining a five man midfield, Noble given license to support lone frontman Lee with Roberts and Matt Richards operating on the flanks.
A misunderstanding between De Vos and Sylvain Legwinski let in Gray who lifted the ball over Supple but Naylor was on hand to head to safety.
Jones then had a gilt edged chance to put the home side ahead after Gray flicked the ball on but the former Crewe striker dragged his shot wide.
Jones was then caught by De Vos, the Town skipper joining Bruce in referee Phil Joslin's book.
Burnley enjoyed most of the possession in the early exchanges of the second-half and Naylor had to react quickly to thwart Jon Harley as he looked to get on the end of O'Connor's quick free-kick.
Jones continued to become an increasingly effective weapon for Burnley, running at the yellow carded Bruce at every opportunity and testing Supple again from distance.
Sito replaced Bruce just before the hour mark, with Lee then picking up his third booking of the season for deliberate handball.
Lee forced a corner for Blues on a break, his shot deflected behind, Frank Sinclair heading clear from Richards' flag kick.
Walton then blazed a strike at goal from 25 yards that saw Jensen making good ground to divert the ball around for a corner as Blues began to enjoy some possession around the home goal.
The travelling Town fans breathed a sigh of relief 17 minutes from time when Jones went down in the box under Richards' challenge but referee Joslin wasn't interested in any penalty appeals.
The Town fans were on their feet soon after with the ball nestling in Burnley's net but Roberts was adjudged offside.
Former Blues' loanee Mahon came on for Elliott for the last 10 minutes as Clarets' boss Steve Cotterill juggled with his line-up, the Republic of Ireland immediately in the action, flashing a cross behind the Town defence and wide of the far post.
John Spicer replaced Jones for the final five minutes, with Kyle Lafferty also coming on for Gifton Noel-Williams.
Town continued to create openings though and a tired Noble almost got on the end of Lee's pass while at the other end De Vos cleared from a Sinclair cross.
With the game entering added on time, McCann won the game for the Clarets with a header from Mahon's corner. The teenage defender left all alone to head in from three yards.
There was still time for Hyde to receive a second yellow and then a red card but Town had no time to make the extra man pay.
